Research and Development of China Instrument Network Instrumentation Researchers at the Institute of Medical Physics, Institute of Modern Physics, Chinese Academy of Sciences, used the conditions provided by the National Laboratory of Heavy Ion Accelerators in Lanzhou to synthesize reduced oxygen sensitizers in the study of the effects of radiation sensitization on gold nanoparticles. The TPZs-AuNPs (TPZs-AuNPs) coupled to TPZ and AuNPs (shown in the figure) were used to study the sensitization effects and mechanisms of TPZs-AuNPs.

The researchers found that the TPZs-AuNPs could enhance the lethal effect of radiation on human hepatoma HepG2 cells; at the same time, the same culture concentration of non-TPZ-coupled gold nanoparticles did not significantly enhance the radiation of HepG2 cells. TPZ plays an important role in the radiation sensitization process.

In the mechanism exploration, the researchers found that under the action of radiation, TPZs-AuNPs can increase the production of hydroxyl radical (·OH) and the level of intracellular reactive oxygen species (ROS) in the simulated solution. In addition, the γ-H2AX foci counts formed on irradiated HepG2 cells also demonstrated that TPZs-AuNPs can enhance the formation of DNA double-strand breaks (DSBs). The researchers concluded that the free radical cascade induced by TPZ mediates the secondary electrons generated by the irradiated gold nanoparticles and is the key mechanism for the radiation sensitization of the TPZs-AuNPs nanoparticles.

These studies confirm that TPZs-AuNPs, as novel gold nanoparticles, are expected to play an important role in radiation sensitization and improve the efficacy of tumor radiotherapy.
